Les souris Spot qui sont un modèle pour le syndrome de Waardenburg couplé à la maladie de
Hirschprung souffrent d’un manque de cellules dérivées de la crête neurale (CCN) incluant les
neurones myentériques ainsi que les mélanocytes de la peau et du vestibule. La paire de gènes
Nr2f1 et A830082k12Rik (K12) est significativement surexprimée dans les CCN entériques
(CCNe) des embryons Spot. De plus, les CCNe des embryons Spot se différencient
prématurément en cellules gliales au détriment des progéniteurs non différenciés. Plusieurs
études récentes suggèrent que de nombreux cas de maladie de Hirschsprung pourraient être
expliqués par une différentiation gliale précoce des CCNe induite par Sonic Hedgehog (SHH).
Notre premier objectif est donc de déterminer si et comment le facteur de transcription Nr2f1
et son paralogue Nr2f2 sont impliqués dans la gliogenèse induite par SHH. Notre second
objectif est de déterminer si le lncRNA K12 est capable de réguler un gène voisin en cis. Nous
avons observé que le traitement de cellules P19 (modèle de cellules souches embryonnaires de
souris) avec SHH induit effectivement l’expression de Nr2f1 et Nr2f2 (mesurée par RT-PCR)
ainsi que celle de plusieurs marqueurs gliaux. De plus, le knock-down de Nr2f1/2 dans les
CCN entériques semble effectivement bloquer la gliogenèse entérique. D’autre part, nos
premières analyses du lncRNA A830082k12Rik montre qu’il est capable de réguler la
transcription d’un gène voisin en cis. Le facteur de transcription NR2F1 semble être un facteur
clé dans la gliogenèse entérique. Une meilleure compréhension de ses mécanismes d’action
ainsi que des voies de signalisation impliqués dans sa régulation semble donc indispensable
pour mieux comprendre cette étape clé du développement du système nerveux entérique. The Spot mouse line is a model for Waardenburg syndrome associated with Hirschsprung’s
disease (Waardenburg syndrome type 4). Spot mice suffer from a lack of neural crest cell
(NCC)-derived cells including myenteric neurons in the colon and melanocytes in the skin and
vestibule. The Nr2f1-A830082K12Rik (K12) gene pair is significantly overexpressed in enteric
NCC (ENCC) of Spot embryos. In addition, ENCC of Spot embryos prematurely differentiate
into glial cells at the expense of undifferentiated progenitors. Several recent studies suggest that
some cases of Hirschsprung’s disease could be explained by an early glial differentiation of
ENCC induced by Sonic Hedgehog (SHH). Our first goal is therefore to determine if and how
NR2F1 transcription factor and its paralog NR2F2 are involved in the SHH-induced gliogenesis.
Our second goal is to determine if the lncRNA K12 is able to regulate a neighboring gene in cis.
We have observed that treatment of P19 cells (mouse embryonic stem cell model) with SHH
effectively induces the expression of Nr2f1 and Nr2f2 (measured by RT-PCR) as well as several
glial markers. In addition, knockdown of Nr2f1/2 in ENCC appears to effectively block enteric
gliogenesis. On the other hand, our first analysis of K12 shows that this lncRNA is able to
regulate the transcription of a neighboring gene in cis. NR2F1 transcription factor appears to be
a key factor in enteric gliogenesis. A better understanding of its mechanisms of action as well
as the signaling pathways involved in its regulation therefore seems essential to better
understand this key stage of the development of the enteric nervous system.
