dcterms.abstract | La nutrition parentérale (NP) est essentielle pour des enfants ayant des troubles intestinaux sévères. Toutefois, elle est associée à des effets secondaires importants. Les peroxydes contaminants la NP limitent l’utilisation de la méthionine comme précurseur de cystéine. Une déficience en cystéine est suggérée par un niveau de glutathion abaissé, limitant la détoxification des peroxydes, et contribuant davantage au stress oxydant. Hypothèse : La déficience en cystéine induite par la NP conduit à une synthèse protéique sous-optimale, particulièrement chez des enfants en croissance. Objectif : Démontrer chez l’animal qu’une supplémentation de la NP en glutathion (GSSG) en tant que procystéine, facilite la synthèse protéique et réduit le stress oxydant. Méthodologie : Des cochons d’Inde d’un mois de vie étaient nourris exclusivement par NP+/-GSSG via un cathéter jugulaire. Un groupe d’animaux nourris avec de la nourriture régulière servit de témoins. Au troisième jour de NP, de la leucine tritiée fut infusée pour évaluer la synthèse protéique. Au cinquième jour, l’hémoglobine, l’urée, le glutathion dans différents organes et l’incorporation protéique au muscle ont été déterminés. Résultats : L’incorporation protéique de radioactivité était de 57% plus élevée chez les NP+GSSG (p<0,05). La concentration réduite en protéines plasmatiques chez le groupe NP (p<0,01) fut élevée au niveau des témoins par l’ajout de GSSG. La présence de GSSG dans la NP a prévenu l’augmentation des marqueurs de stress oxydant observée chez le groupe NP (p<0,01). Conclusion : Enrichir la NP en glutathion pour prévenir une déficience en cystéine représente une méthode simple pour maintenir la synthèse protéique et réduire le stress oxydant chez les individus en croissance. | fr |
dcterms.abstract | Parenteral nutrition (PN) is essential for children with severe intestinal disorders. However, this mode of nutrition is associated with significant side effects. Peroxides contaminating PN limit the use of methionine as precursor of cysteine. Cysteine deficiency is suggested by a lowered level of glutathione, limiting the detoxification of peroxides, contributing even more to oxidative stress. Hypothesis: PN-induced cysteine deficiency leads to suboptimal protein synthesis, especially in growing children. Objective: To demonstrate in an animal model of PN, that glutathione supplementation (GSSG), used as pro-cysteine, facilitates protein synthesis and reduces oxidative stress. Methodology: One-month-old Guinea pigs were exclusively fed by PN+/- GSSG via a jugular catheter. A group of animals fed regular food served as controls. On the third day of PN, tritiated leucine was infused to evaluate protein synthesis. On the fifth day, haemoglobin, urea, glutathione in different organs and protein incorporation into the gastrocnemius muscle were determined. Results: Muscle protein incorporation of tritiated leucine was 57% higher in animals with PN + GSSG (p <0.05). The reduced plasma protein concentration in the PN group (p <0.01) was back to the controls by the addition of GSSG. The presence of GSSG in PN prevented the increase of oxidative stress markers observed in the PN group (p <0.01). Conclusion: In a growing individual, enriching PN with glutathione is a simple way to maintain protein synthesis and reduce oxidative stress by impeding cysteine deficiency. | fr |
